It is typical when reading Edgar Allan Poe’s literature for it to give off a spine-chilling sensation. Poe likes to specify the words he places into his writing, in order for the reader to picture it in their mind. When Poe describes to the reader about the seven rooms he speaks about each color and the glass windows that match the room. He spends a lot of time describing the black room, how it has â€Å"scarlet –a deep blood color† windows. Poe wants the reader to sense that something is not right in the black room â€Å"the dark hangings through the blood-tinted panes, was ghastly in the extreme, and produced so wild a look upon the countenances of those who entered, that there were few of company bold enough to set foot†. He wants one to catch sight of how frightening the view of this room is and how no one set foot inside. He makes the reader feel the suspense of the story as if they are really there. He speaks about the gigantic ebony clock and how it struck hour by hour, and when it did the emotion that went around the room â€Å"the giddiest grew pale, and the more aged and sedate passed their hands over their brows as if in confused reverie or meditation. This makes one feel the intensity of the clocks strikes how Death is an hour closer each strike. These are only a few examples of how Edgar Allan Poe uses diction and imagery in his story â€Å"The Masque of the Red Death† to set a dark and sinister tone. Poe wants the reader to catch the concept of every word he says and why he says it. He also wants one to really feel the hair-rising sensations of the suspense throughout his story.

Phosphate-Buffered Saline or PBS Solution PBS or phosphate-buffered saline is a buffer solution that is particularly valuable because it mimic the ion concentration, osmolarity, and pH of human body fluids. In other words, its isotonic to human solutions, so its less likely to cause cell damage, toxicity,  or unwanted precipitation in biological, medical, or biochemical research. PBS Chemical Composition There are several recipes to prepare PBS solution. The essential solution contains water, sodium hydrogen phosphate, and sodium chloride. Some preparations contain potassium chloride and potassium dihydrogen phosphate. EDTA may also be added in cellular preparation to prevent clumping. Phosphate-buffered saline is not ideal for use in solutions that contain divalent cations (Fe2, Zn2) because precipitation may  occur. However, some PBS solutions do contain calcium or magnesium. Also, keep in mind phosphate may inhibit enzymatic reactions. Be particularly aware of this potential disadvantage when working with DNA. While PBS is excellent for physiological science, be aware the phosphate in a PBS-buffered sample may precipitate if the sample is mixed with ethanol. A typical chemical composition of 1X PBS has a final concentration of  10 mM PO43−, 137 mM NaCl, and 2.7 mM KCl. Heres the final concentration of reagents in the solution: Salt Concentration (mmol/L) Concentration (g/L) NaCl 137 8.0 KCl 2.7 0.2 Na2HPO4 10 1.42 KH2PO4 1.8 0.24 Protocol  for Making Phosphate-Buffered Saline Depending on your purpose, you may prepare 1X, 5X, or 10X PBS. Many people simply purchase PBS buffer tablets, dissolve them in distilled water, and adjust the pH as needed with hydrochloric acid or sodium hydroxide. However, its easy to make the solution from scratch. Here are recipes for 1X and 10X phosphate-buffered saline: Reagent Amountto add (1) Final concentration (1) Amount to add (10) Final concentration (10) NaCl 8 g 137 mM 80 g 1.37 M KCl 0.2 g 2.7 mM 2 g 27 mM Na2HPO4 1.44 g 10 mM 14.4 g 100 mM KH2PO4 0.24 g 1.8 mM 2.4 g 18 mM Optional: CaCl22H2O 0.133 g 1 mM 1.33 g 10 mM MgCl26H2O 0.10 g 0.5 mM 1.0 g 5 mM Dissolve the reagent salts in 800 ml distilled water.Adjust the pH to the desired level with hydrochloric acid. Usually this is 7.4 or 7.2. Use a pH meter to measure the pH, not pH paper or other imprecise technique.Add distilled water to achieve a final volume of 1 liter. Sterilization and Storage of PBS Solution Sterilization isnt necessary for some applications, but if your are sterilizing it, dispense the solution into aliquots and autoclave for 20 minutes at 15 psi (1.05 kg/cm2) or use filter sterilization. Phosphate-buffered saline may be stored at room temperature. It may also be refrigerated, but 5X and 10X solution may precipitate when cooled. If you must chill a concentrated solution, first store it at room temperature until you are certain the salts have completely dissolved. If precipitation does occur, warming the temperature will bring them back into solution. Shelf life of refrigerated solution is 1 month. Diluting a 10X Solution to Make 1X PBS 10X is a concentrated or stock solution, which may be diluted to make a 1X or normal solution. A 5X solution must be diluted 5 times to make a normal dilution, while a 10X solution must be diluted 10 times. To prepare a 1 liter working solution of 1X PBS from a 10X PBS solution, add 100 ml of the 10X solution to 900 ml of water. This only changes the concentration of the solution, not the gram or molar amount of the reagents. The pH should be unaffected.   PBS Versus DPBS Another popular buffer solution is Dulbeccos phosphate buffered saline or DPBS. DPBS, like PBS, is used for biological research and buffers in the 7.2 to 7.6 pH range. It can be stored at room temperature. Dulbeccos solution contains a lower concentration of phosphate. It is 8.1 mM mM phosphate ions, while regular PBS is 10 mM phosphate.  The recipe for 1x DPBS is: Reagent Amount to add (1x) NaCl 8.007 g KCl 0.201 g Na2HPO4 1.150 g KH2PO4 0.200 g Optional: CaCl22H2O 0.133 g MgCl26H2O 0.102 g Dissolve the salts in 800 mL of water. From a critic’s point of view, Tucker states that the US government was undiplomatic in launching such a covert attack against Cambodia at a time when there was hope for a peaceful resolu tion. Owen and Kiernan (1) focused on the revelation of the Cambodia bomb attack after Bill Clinton, the US president released information that the US had concealed about the bombing. Decision making on workplace Essay In my workplace I look after a gentleman who likes to go to the shops on his own. He is deemed too have mental capacity to make this decision. On making this decision it was discussed with him what certain protocols we needed to put in place to support him and to make this happen safely. This man is very much involved in his care plan and he has read, and reviews his care plan when changes are needed. Firstly it was explained to this man that we needed to put a risk assessment in place which says if anything was to happen to him when he was out and about that he took full responsibility of what might happen, for example if this man was to fall when out by himself that the workplace would take no responsibility for this, this man was very happy with this statement and was willing to sign and agree to this risk assessment. The family were also made aware of this decision and were happy for this to be put in place. Family suggested that maybe before he goes out that he was to tell a member of staff where and when he was going, just so that someone is aware. All parties agreed to this. Letting this man go out to the shops by himself really helps with his self-esteem because it makes him feel important, respected and makes him feel like he is as independent as possible and that he doesn’t have to ask all the time about what he wants to do. Also if any changes have been made to his care plan for whatever reason he is always consulted first and asked if he is happy with the decisions that are being made. If he isn’t happy for whatever reason he is more than capable to speak up and say what he would like or prefer. On the other hand if this man didn’t have the mental capacity to make any of these decision or had no family to make decisions on behalf of him and advocate would be brought in and they would be able to make best interest decisions on behalf of him.

I think that, while the boys experience immense bad luck due to the author, the story still proves its point. It is still possible though, that the bad luck of the boys could have been experienced in real life. I think that without this bad luck, the point of the story wouldn’t be as great, because without the restraint’s Golding placed on the boys, life on the island would have been too easy for the boys. The major constraint that Golding puts on the boys is the personality clash between Jack and Ralph. From the beginning, when Ralph is elected leader, Jack hates Ralph, and towards the end of the book, the feeling becomes mutual. Without Jack and Ralph’s problems, life would have been easy, and the ‘darkness of man’s heart’ would not have been conveyed to the reader. Jack shows ‘the darkness’ and if he and Ralph had just been friends, there would never have been an opportunity for Jack to show this darkness which lurked beneath the surface. Golding also uses the dead pilot conveniently against the boys - the way in which he is caught in the trees just in the right position to be caught by the wind and look like the beast and the way the wind picks up after Simon has let him down from the trees and carries him out to sea, so that the other boys cannot see that it wasn’t a beast. The author uses the boy’s fear against them, and although this could possibly happen in the situation, Golding uses it as a weapon against them, their morale and their companionship. I think that the boys split up and go to Jack because of the fear - he can kill the beast, he can get them meat, and if they ever get upset, he can start a dance and all will be fine.

A Walk To Remember Compare Amp Contrast

Both involve a Story Of a boy and a girl, almost completely different in their views, which slowly fall in love n Barefoot, North Carolina. Throughout the movie the struggles of the characters are awfully similar to the struggles of the characters in the book, with a few changes here and there. Although the book and movie are similar, there are still differences between the two such as how the two got to know each other, how the characters are portrayed, as well as how certain events occurred and how the characters reacted to them.The first major difference between the novel and the movie of A Walk to Remember is how different the characters are portrayed in each. For example, throughout the novel the leading girl, Jamie Sullivan, was the shy daughter of the towns Pastor. She was one of the kindest girls anyone had met, she would help anyone whether she was asked or not. She had a big part in the town's orphanage, and she had worshiped God more than anyone had seen. Jamie was an inte lligent conservative girl who wore her blonde hair put in a tight bun all the time.She wasn't known to have many friends, or even do ‘normal' teenager things, and she usually spent her time at school, at the orphanage, or with her father. In comparison, in the movie, Jamie was quite similar to these qualities; she was smart, very religious, shy, kind, and conservative showed by how she had dressed. However, even though Jamie was kind and seemed to be gentle, in the movie it didn't portray how religious she actually was as well as I thought they could've. In addition, don't believe that it showed how much she actually cared for others and wished to help them.For example, Jamie would help younger kids and tutor and do things like that, but in the novel she also worked at an orphanage and would even elf little animals in the middle of the road, but in the movie you didn't truly see that side of Jamie as much. Now on the other hand, the main character, Logan Carter, is quite diffe rent in the movie than how he was described in the novel. F-or example, the very first scene of the movie is Logan playing a mean trick with his friends on a boy.In this scene you can already tell that Logan is supposed to be a â€Å"bad boy' and that h?s supposed to be trouble. In the novel Logan was somewhat of a trouble maker when he was younger, which is possibly what gave the writers the idea to make him a bad boy in the movie. But in reality, Logan is just your average confused teenage boy. He likes to hang out with his friends, go on dates, and didn't enjoy school all that much. Still, in the movie it portrays him as being careless and self centered until Jamie made him realize that he was making some bad choices.Another difference between the novel and the book of A Walk to Remember is how Jamie and Logan actually got to know each other. For example, in the novel they first become a little closer after Logan asks Jamie to the homecoming dance, only because he needed a date and she was the only one aft. But in the movie, they both get cast to do the annual town play, which does happen in the book but it wasn't their first encounter. As the story goes on Logan, as a punishment for his prank in the beginning of the movie, he must participate in the play which he ends up getting the lead role in.He starts to care a little more about the play as the movie progresses and he practices his lines more and more, and eventually asks Jamie to help him with his lines and she agrees to help but on one condition â€Å"he has to promise not to fall in love with her†. Now that line may be cute and seem not that important, but its an incredibly significant quote to the story in my opinion. This quote was used in the novel as well when London had asked Jamie to go to the homecoming dance with her, which I believe shows a little importance that the movie writers had kept such a specific quote.In addition to that, a considerable difference from the book to the movi e would, in my opinion, be the fact that in the novel they fell in love slowly and hung out at one another's house, while in the movie they fell in love faster and did more ‘romantic' or public outings. Also Logan seemed to do more things to â€Å"win† her over in the book such as him visiting the orphanage with her and getting a great deal of money without her knowledge so she could get the orphans Christmas presents, while in the movie they both seemed to just fall in love with each other.Lastly, certain events and reactions differed from the book to the movie. For instance, in the book Lagan's friends would make fun of and tease him for his attraction to Jamie and for spending time and being friends with her. But they never would have dared make fun of Jamie to her face, mainly because they let like she was on Gods â€Å"good side† or had it â€Å"in† with God. Even if they had made fun of her to her face you could tell, just from what you have read tha t she wouldn't have done anything about it and she wouldn't have had a comeback for them.But on the contrary, in the movie Logan and his friends would make fun of her to her face and ridicule her. Even though she was supposed to be Godly and kind, she still remained above it all and even had some types of comebacks. For example, in one scene one Of Lagans friends had said â€Å"If there is a higher power, why is it He can't get you a new sweater? And Jamie replies, â€Å"Because He's too busy looking for your brain†.The last difference between the book and the movie is that in the book when Logan found out about Jamie illness he felt uncomfortable and didn't know what to do. But on the other hand in the movie he seemed to be devoted to her and would do anything for her and he knew exactly what to do, even if he was in denial for a little bit after he heard the news. To conclude, there are differences between the book and the movie, some may be minor some more noticeable, but overall the book and the movie are Mathew similar to each other.

The motorcycle can also be a disadvantage because while you may be able to drive farther per gallon of fuel, one must refill the tank more often. A motorcycle can travel approximately 220 miles before needing more fuel, but a car can go almost 500 miles. This can be an advantage for someone who likes to drive long distances without having to stop for fuel very often. While the 3 improvements to a cars safety systems have improved every year, the safety of riding a motorcycle have remained unchanged. The disadvantage of riding a motorcycle is that the only safety devices the rider has is in the gear they choose to wear. Unfortunately, even if the rider wears everything, it is almost certain that the rider will be severely injured or killed in a major accident. The last thing to think about is the passenger and storage capabilities of the vehicle Having the capability to carry both passengers and cargo is the last and most important determining factor in the decision of buying a car or a motorcycle. A car has the advantage if the person has a family or intends on carrying passengers, with the capability to carry up to five passengers, where a motorcycle can only carry one. The cargo carrying capability of a motorcycle is extremely limited to the size of containers that need to be bought separately and attached to the motorcycle.
