It is typical when reading Edgar Allan Poeââ¬â¢s literature for it to give off a spine-chilling sensation. Poe likes to specify the words he places into his writing, in order for the reader to picture it in their mind. To make this possible, Edgar Allan Poe utilizes negative diction and imagery to manifest a dark and sinister tone in his story, ââ¬Å"The Masque of the Red Deathâ⬠. Diction is the word choice a writer uses in their writing to develop a special meaning. Poe wants to send a dark emotion to his readers and starts off his story by personifying ââ¬Å"Deathâ⬠, giving it human qualities, portraying it is as the protagonist. He wants to make sure the reader realizes how important Death is in the story. Poe also speaks a lot about death and the horrible way these people are dying. This gives off a sinister tone of how he constantly refers to death. When Poe describes to the reader about the seven rooms he speaks about each color and the glass windows that match the room. He spends a lot of time describing the black room, how it has ââ¬Å"scarlet ââ¬âa deep blood colorâ⬠windows. Poe wants the reader to sense that something is not right in the black room ââ¬Å"the dark hangings through the blood-tinted panes, was ghastly in the extreme, and produced so wild a look upon the countenances of those who entered, that there were few of company bold enough to set footâ⬠. He wants one to catch sight of how frightening the view of this room is and how no one set foot inside. He makes the reader feel the suspense of the story as if they are really there. He speaks about the gigantic ebony clock and how it struck hour by hour, and when it did the emotion that went around the room ââ¬Å"the giddiest grew pale, and the more aged and sedate passed their hands over their brows as if in confused reverie or meditation. This makes one feel the intensity of the clocks strikes how Death is an hour closer each strike. These are only a few examples of how Edgar Allan Poe uses diction and imagery in his story ââ¬Å"The Masque of the Red Deathâ⬠to set a dark and sinister tone. Phosphate-Buffered Saline or PBS Solution PBS or phosphate-buffered saline is a buffer solution that is particularly valuable because it mimic the ion concentration, osmolarity, and pH of human body fluids. In other words, its isotonic to human solutions, so its less likely to cause cell damage, toxicity,à or unwanted precipitation in biological, medical, or biochemical research. PBS Chemical Composition There are several recipes to prepare PBS solution. The essential solution contains water, sodium hydrogen phosphate, and sodium chloride. Some preparations contain potassium chloride and potassium dihydrogen phosphate. EDTA may also be added in cellular preparation to prevent clumping. Phosphate-buffered saline is not ideal for use in solutions that contain divalent cations (Fe2, Zn2) because precipitation mayà occur. However, some PBS solutions do contain calcium or magnesium. Also, keep in mind phosphate may inhibit enzymatic reactions. Be particularly aware of this potential disadvantage when working with DNA. While PBS is excellent for physiological science, be aware the phosphate in a PBS-buffered sample may precipitate if the sample is mixed with ethanol. A typical chemical composition of 1X PBS has a final concentration ofà 10 mM PO43âËâ, 137 mM NaCl, and 2.7 mM KCl. Heres the final concentration of reagents in the solution: Salt Concentration (mmol/L) Concentration (g/L) NaCl 137 8.0 KCl 2.7 0.2 Na2HPO4 10 1.42 KH2PO4 1.8 0.24 Protocolà for Making Phosphate-Buffered Saline Depending on your purpose, you may prepare 1X, 5X, or 10X PBS. Many people simply purchase PBS buffer tablets, dissolve them in distilled water, and adjust the pH as needed with hydrochloric acid or sodium hydroxide. However, its easy to make the solution from scratch. Here are recipes for 1X and 10X phosphate-buffered saline: Reagent Amountto add (1) Final concentration (1) Amount to add (10) Final concentration (10) NaCl 8 g 137 mM 80 g 1.37 M KCl 0.2 g 2.7 mM 2 g 27 mM Na2HPO4 1.44 g 10 mM 14.4 g 100 mM KH2PO4 0.24 g 1.8 mM 2.4 g 18 mM Optional: CaCl22H2O 0.133 g 1 mM 1.33 g 10 mM MgCl26H2O 0.10 g 0.5 mM 1.0 g 5 mM Dissolve the reagent salts in 800 ml distilled water.Adjust the pH to the desired level with hydrochloric acid. Usually this is 7.4 or 7.2. Use a pH meter to measure the pH, not pH paper or other imprecise technique.Add distilled water to achieve a final volume of 1 liter. Sterilization and Storage of PBS Solution Sterilization isnt necessary for some applications, but if your are sterilizing it, dispense the solution into aliquots and autoclave for 20 minutes at 15 psi (1.05 kg/cm2) or use filter sterilization. Phosphate-buffered saline may be stored at room temperature. It may also be refrigerated, but 5X and 10X solution may precipitate when cooled. If you must chill a concentrated solution, first store it at room temperature until you are certain the salts have completely dissolved. If precipitation does occur, warming the temperature will bring them back into solution. Shelf life of refrigerated solution is 1 month. Diluting a 10X Solution to Make 1X PBS 10X is a concentrated or stock solution, which may be diluted to make a 1X or normal solution. A 5X solution must be diluted 5 times to make a normal dilution, while a 10X solution must be diluted 10 times. To prepare a 1 liter working solution of 1X PBS from a 10X PBS solution, add 100 ml of the 10X solution to 900 ml of water. This only changes the concentration of the solution, not the gram or molar amount of the reagents. The pH should be unaffected.à PBS Versus DPBS Another popular buffer solution is Dulbeccos phosphate buffered saline or DPBS. DPBS, like PBS, is used for biological research and buffers in the 7.2 to 7.6 pH range. It can be stored at room temperature. Dulbeccos solution contains a lower concentration of phosphate. It is 8.1 mM mM phosphate ions, while regular PBS is 10 mM phosphate.à The recipe for 1x DPBS is: Reagent Amount to add (1x) NaCl 8.007 g KCl 0.201 g Na2HPO4 1.150 g KH2PO4 0.200 g Optional: CaCl22H2O 0.133 g MgCl26H2O 0.102 g Dissolve the salts in 800 mL of water. According to the VVAW (Para. 1), the Cambodia bombing continued despite a peace treaty ââ¬Å"bombing haltâ⬠that required that the US stop its attack in Indo-China. The US launched over 500 thousand tons of bombs. The main targets of the bombs were the peopleââ¬â¢s army of Vietnam (PAVN) and forces of the Viet Cong and their air bases that were used to as the supply and training areas. The US persisted bombing despite the countries uncooperativeness to the diplomatic missions launched to end their conflict with Vietnam. According toTucker (740), this was a war whose intentions were personal and did not rhyme with the international strategies of resolving conflicts. This is the reason why the US kept the mission secret because they did not have any good reason to attack this country. From a criticââ¬â¢s point of view, Tucker states that the US government was undiplomatic in launching such a covert attack against Cambodia at a time when there was hope for a peaceful resolu tion. Other scholars have provided that the US president Nixon depicted the highest level of imperialism in their attack of the Cambodia. Owen and Kiernan (1) focused on the revelation of the Cambodia bomb attack after Bill Clinton, the US president released information that the US had concealed about the bombing. The purpose of Bill Clinton was to show the USââ¬â¢s accountability and its commitment to establish honesty in its history. Decision making on workplace Essay In my workplace I look after a gentleman who likes to go to the shops on his own. He is deemed too have mental capacity to make this decision. On making this decision it was discussed with him what certain protocols we needed to put in place to support him and to make this happen safely. This man is very much involved in his care plan and he has read, and reviews his care plan when changes are needed. Firstly it was explained to this man that we needed to put a risk assessment in place which says if anything was to happen to him when he was out and about that he took full responsibility of what might happen, for example if this man was to fall when out by himself that the workplace would take no responsibility for this, this man was very happy with this statement and was willing to sign and agree to this risk assessment. The family were also made aware of this decision and were happy for this to be put in place. Family suggested that maybe before he goes out that he was to tell a member of staff where and when he was going, just so that someone is aware. All parties agreed to this. Letting this man go out to the shops by himself really helps with his self-esteem because it makes him feel important, respected and makes him feel like he is as independent as possible and that he doesnââ¬â¢t have to ask all the time about what he wants to do. Also if any changes have been made to his care plan for whatever reason he is always consulted first and asked if he is happy with the decisions that are being made. If he isnââ¬â¢t happy for whatever reason he is more than capable to speak up and say what he would like or prefer. On the other hand if this man didnââ¬â¢t have the mental capacity to make any of these decision or had no family to make decisions on behalf of him and advocate would be brought in and they would be able to make best interest decisions on behalf of him.

Lord of the Flies Artificial Restraints in Lord of the Flies "GOLDING PUTS SO MANY ARTIFICIAL RESTRAINTS ON HIS STORY IN ORDER TO EMPHASISE HIS POINT, THAT THE WHOLE THING COMES OUT TOO NEATLY AND, IN FACT, REDUCES THE POWER OF HIS MESSAGE." I think that, while the boys experience immense bad luck due to the author, the story still proves its point. It is still possible though, that the bad luck of the boys could have been experienced in real life. I think that without this bad luck, the point of the story wouldnââ¬â¢t be as great, because without the restraintââ¬â¢s Golding placed on the boys, life on the island would have been too easy for the boys. The major constraint that Golding puts on the boys is the personality clash between Jack and Ralph. From the beginning, when Ralph is elected leader, Jack hates Ralph, and towards the end of the book, the feeling becomes mutual. Without Jack and Ralphââ¬â¢s problems, life would have been easy, and the ââ¬Ëdarkness of manââ¬â¢s heartââ¬â¢ would not have been conveyed to the reader. Jack shows ââ¬Ëthe darknessââ¬â¢ and if he and Ralph had just been friends, there would never have been an opportunity for Jack to show this darkness which lurked beneath the surface. Golding also uses the dead pilot conveniently against the boys - the way in which he is caught in the trees just in the right position to be caught by the wind and look like the beast and the way the wind picks up after Simon has let him down from the trees and carries him out to sea, so that the other boys cannot see that it wasnââ¬â¢t a beast. The author uses the boyââ¬â¢s fear against them, and although this could possibly happen in the situation, Golding uses it as a weapon against them, their morale and their companionship. I think that the boys split up and go to Jack because of the fear - he can kill the beast, he can get them meat, and if they ever get upset, he can start a dance and all will be fine.
